Yeast One-Hybrid (Y1H) System Service
The yeast one-hybrid system is a technique used to study the interaction between DNA and protein. Its characteristic is that it can identify proteins that are stably bound to DNA, can study the DNA-protein interactions in eukaryotes in yeast cells, and directly obtain the coding genes of target sequence interaction proteins by screening DNA libraries.
The principle of the yeast one-hybrid system is to construct a known specific cis-acting element structure upstream of the most basic promoter, and construct a reporter gene downstream of it. Then the fusion expression vector of the cDNA encoding the transcription factor to be tested and the known yeast transcription activation domain is introduced into yeast cells. If the gene can be combined with the cis-acting element, the most basic promoter can be activated and the reporter gene can be expressed.
