Transcription Factors (TFs) are proteins that can bind DNA in a sequence-specific manner and regulate transcription. Transcription factors control chromatin and transcription by recognizing specific DNA sequences to form a complex system that directs genome expression. The experimental principle of the system is to introduce the method of sequence randomization to construct a random DNA library, and then use the yeast one-hybrid technology to screen the library to identify the DNA sequence that specifically binds to the transcription factor. The transcription factor studied is Bait, and a random short DNA sequence library is cloned into vector as Prey. One element sequence is enough to bind to TF.
