Prokaryotic Yeast One-Hybrid (Y1H) Service
Yeast one-hybrid technology is developed from the yeast two-hybrid technology. Through the phenotype detection of the reporter gene, the interaction between DNA and protein is analyzed to study the regulation of gene expression in eukaryotic cells. Due to the sensitivity and reliability of the yeast one-hybrid method to detect specific interactions between specific transcription factors and cis-acting elements, it has been widely used in cloned cells that contain weak specific transcription factors that are difficult to purify by biochemical means.
The theoretical basis of yeast one-hybrid (Y1H) is that the transcription activator of many eukaryotes is composed of DNA binding region (BD) and transcription activation region (AD), which are physically and functionally independent of each other. Therefore, a fusion expression vector of various genes and AD can be constructed. When expressed as a fusion protein in yeast, a protein with a specific binding region to the target element can be screened based on the expression of the reporter gene. Theoretically, in one-hybrid detection, any target element can be used to screen for a protein with a specific binding region.
Service introduction
Traditional yeast one-hybrid library construction methods are mostly for eukaryotes, because eukaryotic mRNA has a polyA structure. However, because bacterial prokaryotes do not have a polyA structure, it is difficult to construct a yeast one-hybrid library of bacterial prokaryotes using traditional methods. Creative BioMart has developed a special prokaryotic library construction and screening method for bacterial prokaryotes, which can stably, quickly and efficiently construct a high-quality yeast one-hybrid library of prokaryotic samples.
