Genomic Yeast One-Hybrid (Y1H) Service
Yeast one-hybrid is an effective method for cloning transcription factor genes (cDNA) that specifically bind to target elements based on the principle that DNA binding proteins bind to DNA cis-acting elements to regulate the expression of reporter genes. The theoretical basis is that many eukaryotic transcription activators are composed of physically and functionally independent DNA-binding domains (BD) and transcriptional activation domains (AD). Therefore, various genes and AD can be constructed. When the fusion expression vector is expressed as a fusion protein in yeast, according to the expression of the reporter gene, the protein that has a specific binding region with the target element can be screened out. Theoretically, in one-hybrid detection, any target element can be used to screen for a protein with a specific binding region.
