Yeast Two-Hybrid (Y2H) System Service
The establishment of the yeast two-hybrid system (Y2H) is based on the understanding of eukaryotic regulation of transcription initiation. Initiation of gene transcription by cells requires the participation of trans-activator. The yeast two-hybrid system is to clone (fusion) the two proteins to be studied into the DNA binding domain (DNA-BD) and transcription activation domain (AD) of the transcription activator (such as modular GAL4, etc.) of the yeast expression plasmid. Above, it is constructed into a fusion expression vector, and the system for analyzing the interaction of two proteins from the expression product. When the two binding domains are separated, they still function separately, but they cannot activate transcription. When the two are relatively close in space through appropriate pathways, the GAL4 transcription factor activity can be reproduced, and the downstream promoter of the upstream activation sequence (UAS) can be activated, so that the downstream genes of the promoter can be transcribed.
In the yeast two-hybrid system, the "bait" protein X is cloned into the DNA-BD vector to express the DNA-BD/X fusion protein; the protein Y to be tested is cloned into the AD vector to express the AD/Y fusion protein. Once there is an interaction between the X and Y proteins, DNA-BD and AD are also drawn closer, resume their functions, and activate the expression of LacZ and HIS3 genes in the reporter recombinant.
