Yeast Two-Hybrid (Y2H) Library Screening Service
The protein yeast two-hybrid experiment is based on the genetic analysis of yeast, an experiment to study the influence of the interaction between trans-acting factors on the regulation of eukaryotic gene transcription. Transcription activating proteins can bind to specific sequences on DNA to initiate the transcription response of corresponding genes. This function of DNA binding and transcription activation is performed by two independent domains on transcription activation protein, namely DNA binding domain (BD) and transcription activation domain (AD). And these two domains are necessary for the transcriptional activation of genes.

The systems currently used in yeast two-hybrid experiments include LexA system and Gal4 system. In the LexA system, the DNA binding domain is composed of a complete prokaryotic protein LexA, and the transcription activation domain is composed of an acidic E. coli polypeptide B42 of 88 amino acids, which can activate gene transcription in yeast; In the Gal4 system, BD and AD are respectively composed of two different domains (1-147aa and 768-881aa) on the Gal4 protein. When using the GAL4 system to screen a cDNA library or study the interaction between proteins, the DNA binding domain binds to the target protein or "bait", and the transcription activation domain binds to the library protein or the protein to be verified.
