Yeast Surface Display Analysis of Glycosylphosphatidylinositol (GPI) Anchoring System
The flocculin display expression system utilizes the flocculin gene to fuse with foreign proteins and express the fusion protein on the cell surface. Its subsystem - GPI anchoring system uses the GPI signal contained in the C-terminus of the flocculin Flo1p to anchor foreign proteins. This system is similar to the agglutinin display system.
Yeast flocculation gene (Flo1) is a gene in the flocculation gene family of Saccharomyces cerevisiae that has been studied in detail, and the yeast surface display system is also operated based on this gene. Flo1 is dominant and encodes a lectin-like protein rich in serine and threonine. From N-terminus to C-terminus, Flo1 protein is composed of secretion signal region, 4-segment repeat sequence region and GPI anchor attachment signal region in sequence.

Technical advantages

- The yeast display system uses FACS sorting technology, based on antibody affinity and display level, which can eliminate the deviation caused by expression and distinguish clones with low affinity.
- By using double-stained FACS, antibody affinity can be measured directly on the surface of yeast cells, so there is no need for time-consuming subcloning, expression and purification.
- In the same display system, the affinity and stability of antibodies can be optimized.
