Yeast Surface Display Analysis of Flocculation Domain Anchoring System
The N-terminal flocculation functional region system of Flo1 protein consists of secretion signal region, Flo1 protein flocculation functional region and target protein insertion site. The flocculation functional domain anchoring system uses the flocculation functional domain in the middle of Flo1p to fuse with the N-terminal of the foreign protein. The flocculation functional domain recognizes the mannan chains assembled in the yeast cell wall, and induces cell adhesion and aggregation into a reversible flocs.

Technical advantages
- Yeast is a eukaryotic organism, with the organelles and enzymes required for post-translational modification, so that the diversity of genes can be accurately displayed through correctly folded proteins, so that expression deviations are small.
- The displayed fusion protein has a wide range of relative molecular mass and a large number of molecules. Studies have found that molecules with relative molecular masses between 0.93×106 Da and 136×106 Da can be displayed on the surface of yeast, and each yeast surface can display 104 to 105 molecules.
- Either traditional methods (binding-elution-amplification screening) or fluorescence activated cell sorter (FACS) can be used for screening.
- The mutation characteristics and enzyme activity of a single clone displayed on the cell surface do not need to be water-soluble expression and purification of the protein, and it can be measured directly on a whole-cell basis, and the activity measurement is convenient.
