Yeast Surface Display Analysis of a-Agglutinin System (Aga)
The cell wall of Saccharomyces cerevisiae is mainly composed of the outer layer of mannoprotein and the inner layer of glucan skeleton, which are connected by covalent bonds. α-agglutinin and a-agglutinin are two mannoproteins on the cell wall of yeast. They mediate adhesion between the haploid cells of the α-mating type (MATα) and the α-mating type (MATa) of Saccharomyces cerevisiae, thereby making the cells fuse to form a diploid.
a-agglutinin is composed of core subunit Aga1 and binding subunit Aga2, which are connected by disulfide bonds. The C-terminus of the core subunit Aga1 is covalently bound to the dextran of the cell wall. The target protein is fused to the C-terminus of the binding subunit Aga2, that is, the N-terminus of the protein itself is fused to the C-terminus of the binding subunit Aga2 of a-agglutinin. Therefore, a reducing agent can be used to break the disulfide bond to dissociate the target protein from the cell surface, which is convenient for separation and purification.
