Yeast CUT&Tag Analysis

CUT&Tag (Cleavage Under Targets and Tagmentation) is a new technique for DNA-protein interaction studies that can be applied in the field of epigenetics, mainly for studying the binding or distribution sites of transcription factors or histone modifications on the whole genome, or for studying the interaction between proteins and DNA and identifying target DNA binding sites of target proteins. The technical objectives are similar to those of Chip-seq.
The core of the technology is hyperactive pG-Tn5 / pA-Tn5 transposase, which is a fusion of protein G/A with an engineered ultra-active Tn5 transposase, resulting in a novel fusion enzyme with both transposase and Protein G/A activity.
CUT & Tag works by first making the cell membrane permeable, then incubating the primary antibody with the target protein, then incubating with the secondary antibody, and finally adding the complex of proteinA-Tn5 and the sequencing junction. (by translocating the DNA around the target protein and ligating the sequencing junction). Finally, the sequencing library will be obtained by PCR.
Yeast needs to break the cell wall or extract the nucleus to perform the experiment.
