Small Molecule Three-Hybrid (Y3H) Service for Drug Target Screening
The small molecule three-hybrid system is a small molecule chemical drug analysis technology based on the yeast two-hybrid (Y2H) technology, which is used for the screening and identification of drug targets. This technology uses small molecules or drugs of interest as bait to screen highly complex protein domain libraries prepared from any type of cell, tissue or organism. Since the active transcription factor is partially reconstructed from the DNA binding domain (DBD) and activation domain (AD), it can be used to identify drug-protein interactions.
The small molecule three-hybrid system uses cell-permeable polymerized small molecule chemical inducers (CIDs) to promote the oligomerization of chimeric receptors in mammalian cells. When the "drug - target protein" interaction occurs, the labeled drug closes the gap between DBD and AD due to the tag-ABD interaction. This makes it possible to express the reporter gene and grow yeast on a selective medium, and to analyze the positive clones by sequencing to identify the target protein.
