Saccharomyces cerevisiae Protein Expression Analysis
Saccharomyces cerevisiae is difficult to culture at high density, and its secretion efficiency is low. It hardly secretes foreign proteins with a molecular weight greater than 30 kD, nor can the expressed foreign proteins be correctly glycosylated, and the C-terminus of expressed proteins is often truncated. Therefore, Saccharomyces cerevisiae is generally not used as the host bacteria for recombinant protein expression.
Saccharomyces cerevisiae itself contains a plasmid, and its expression vector can have two types: autonomous replication type and integration type. Autonomously replicating plasmids usually have 30 or more copies and contain automatic replicating sequences (ARS), which can replicate independently of yeast chromosomes. If there is no selective pressure, these plasmids are often unstable. The integrated plasmid does not contain ARS, it must be integrated into the chromosome and replicate with the chromosome replication. The integration process is highly specific, but the copy number is very low.
To this end, the pMIRY2 (for multiple integration into ribosomal DNA in yeast) plasmid was designed to target the target gene to be integrated into the rDNA cluster (rDNA cluster is the 150 repetitive sequences that exist in tandem in the yeast genome). Therefore, more than 100 copies can be obtained using the pMIRY2 plasmid. It is worth noting that foreign proteins expressed by Saccharomyces cerevisiae are often highly glycosylated, and the sugar chain can have more than 40 mannose residues. The core oligosaccharide chain of glycoprotein contains only 1,3 mannose at the end, and the antigenicity of the product is significantly enhanced. Therefore, Saccharomyces cerevisiae is often used to prepare subunit vaccines (such as HBV vaccine, foot-and-mouth disease vaccine, etc.).
