The Red system of Saccharomyces cerevisiae is used to carry out homologous recombination of foreign DNA to achieve allelic replacement of target genes. Then, antibiotics are used to screen for the insertion mutant strains with the suicide vector integrated into the target site of the bacterial genome. Under the second round of reverse selection pressure, only the second homologous recombination of the Saccharomyces cerevisiae genome and the loss of the suicide plasmid can survive.
