PRM-targeted Proteomic Analysis of Yeast
Parallel Reaction Monitoring (PRM) is an ion monitoring technology based on high-resolution and high-precision mass spectrometry, which can selectively detect target proteins and target peptides, so as to achieve accurate quantification of target proteins and peptides. Targeted proteome scanning is like a precision sniper, eliminating distractions and having a clear target.
Its technical principle is relatively simple. First, a quadrupole mass analyzer with high-resolution, high-precision mass spectrometry (such as the Orbitrap series) is used to selectively collect the target peptide precursorions in the primary mass spectrometry. Subsequently, the precursor ions are fragmented into fragment ions in the collision cell, and all enter the secondary mass spectrometer Orbitrap analyzer, which will produce a high-resolution secondary mass spectrum. Finally, all product ion information of the target peptide is collected to realize the specific analysis of target protein/peptide in complex samples.
