iTRAQ Proteomic Analysis of Yeast
iTRAQ/TMT labeling quantitative proteome research is to label all proteins expressed in a genome or all proteins in a complex mixed system, and use the secondary reporter ions in the labeling reagent to accurately identify and quantify the proteins. There are two kinds of labeling reagents, namely iTRAQ (Isobaric Tag for Relative Absolute Quantitation) and TMT (Tandem Mass Tags), which are peptide labeling and quantitation technologies in vitro. They use tags of multiple isotopes, which can react with amino groups (including N-terminal amino acids and amino groups of lysine side chains) to achieve linkages. Through high-precision mass spectrometry analysis, they can simultaneously identify and quantify multiple sample proteomes.
