Full-length Transcriptome Sequencing and Analysis Service of Yeast
The complete transcript includes the sequence from the 5'end to the 3'end of the polyA tail, and the length is concentrated in 1-6 kb. Due to the short read length of the second-generation sequencing technology, the sequence fragments obtained need to be spliced, and the resulting transcripts may have splicing errors and more chimeras, so that a complete transcript cannot be obtained. However, the third-generation sequencing is based on the Pacbio platform, using single-molecule real-time sequencing technology (SMRT), due to its ultra-long read length (average 15kb), you can directly obtain a complete full-length transcript without splicing. Therefore, the problem of short splicing of transcripts and incomplete information of species without reference genome is overcome, and the study of structural variations such as variable splicing and fusion genes in species with reference genome can be realized.
For yeast without a reference sequence, full-length transcriptome sequencing can construct a high-quality gene set, laying the foundation for subsequent functional studies of yeast. For yeasts with reference sequences, new transcripts can be discovered, reference gene sets can be improved, and structural variations such as alternative splicing and gene fusion can be identified.

Experiment procedure

Sample requirements
- Total RNA ≥ 10 μg, concentration ≥ 300 ng / μl, RIN ≥ 8.0, OD260 / 280 = 1.6~2.2,OD260 / 230= 1.4~2.5
- Yeast cell ≥ 2×107 ~ 2×108
Please put the sample in a 1.5 ml tube, indicate the name, concentration and preparation time of the sample on the tube, and seal the tube with parafilm. Before shipping, fix all sample tubes in 50 ml centrifuge tubes with caps and ship them on dry ice.
