AQUA-targeted Proteomic Analysis of Yeast
Absolute quantitative analysis (AQUA) is a targeted quantitative proteomics technique that is widely used in various quantitative proteomic studies. Absolute quantification strategies can be used for absolute quantification of proteins and their modification states. Using stable isotope-labeled synthetic peptides as internal standards, it can simulate natural peptides formed by proteolysis, or can be prepared by covalent modifications (phosphorylation, methylation, acetylation, etc.). By using selected reaction monitoring (SRM) analysis in tandem mass spectrometry, the AQUA internal standard peptide was used for precise quantitative determination of absolute levels of proteins and post-translationally modified proteins after proteolysis.
