Acetylation Proteomic Analysis of Yeast
Acetylation is a highly conserved and reversible protein modification in vivo, which plays a very important role in the activation of transcriptional regulators in the nucleus. In addition, there are a large number of non-histone acetylation modifications involved in the regulation of metabolic pathways and metabolic enzyme activities. Due to the low content and wide dynamic range of acetylated proteins in biological samples, acetylated peptides need to be enriched before mass spectrometry analysis to improve their abundance, and then traditional quantitative proteome analysis methods are used to analyze the enriched acetylated peptides Quantitative analysis of peptide samples.
Creative BioMart optimized the experimental method, using peptide pre-separation to reduce the impact of high-abundance histones on protein acetylation identification, and then combined with co-immunoprecipitation to enrich acetylated peptides through efficient antibodies, so as to achieve large-scale acetylation identification and quantitative.
